Assay Development Phase (~3-6 months)
- User submits assay plan to OTRADI
- User develops primary screen, with secondary assay in place
- OTRADI works with User to make assay amenable to automation, including miniaturization to 96-well and/or 384-well format
- OTRADI & User create written automation protocol
- OTRADI & User demonstrate assay robustness and suitability for the HTS or HCA format to
- determine when assay metrics are suitable for screening
- OTRADI creates screening schedule with projected start date & timeline
Scaling Assays & Performing Screens (~2-3 months)
- OTRADI coordinates reagents, consumables & compound libraries
- OTRADI performs pilot screen of up to 2000 compounds in one batch
- OTRADI & User analyze pilot screen data. If acceptable, continue timeline to screen additional larger compound libraries
Data-Processing, Cherry Picking & Hit Confirmation (~2-3 months)
- OTRADI creates Assay Report summarizing results of compound library screens & distributes to User
- OTRADI manages QC metrics and hits
- User & OTRADI choose compounds for cherry picking and hit confirmation
Potency & Follow-On Experiments (~2-3 months)
- After hit confirmation, OTRADI & User may schedule dose-response experiments to measure potency of hit
- OTRADI & User can discuss, plan and schedule follow-on experiments (secondary assays, cytotoxicity, cellular analysis (HCA), PK, etc.)
